TGFB-induced factor homeobox 2

TGIF2
The protein encoded by this gene is a DNA-binding homeobox protein and a transcriptional repressor, which appears to repress transcription by recruiting histone deacetylases to TGF beta-responsive genes. This gene is amplified and over-expressed in some ovarian cancers. Alternative splicing results in multiple transcript variants. A related pseudogene has been identified on chromosome 1. Read-through transcription also exists between this gene and the neighboring downstream C20orf24 (chromosome 20 open reading frame 24) gene. [provided by RefSeq, Dec 2010] (from NCBI)

Review
Parini et al., Huddinge, Sweden. In Biochim Biophys Acta, 2015
As these proteins can interact with the ligand binding domain of retinoid X receptor α, a common heterodimeric partner of several nuclear receptors [e.g., liver X receptors (LXRs) and peroxisome proliferator-activated receptors (PPARs)], Tgif1 and Tgif2 might repress other transcriptional pathways activated by lipids.
MicroRNA-34a inhibits tumor invasion and metastasis in gastric cancer by targeting Tgif2.

Role of pyruvate kinase M2 in transcriptional regulation leading to epithelial-mesenchymal transition.
Ishii et al., Ōsaka, Japan. In Proc Natl Acad Sci U S A, 2014
Immunoprecipitation and LC-electrospray ionized TOF MS analyses revealed that EMT stimulation causes direct interaction of PKM2 in the nucleus with TGF-β-induced factor homeobox 2 (TGIF2), a transcriptional cofactor repressor of TGF-β signaling.
miR-34a blocks osteoporosis and bone metastasis by inhibiting osteoclastogenesis and Tgif2.
Impact
Wan et al., Dallas, United States. In Nature, 2014
Mechanistically, we identify transforming growth factor-β-induced factor 2 (Tgif2) as an essential direct miR-34a target that is pro-osteoclastogenic. Tgif2 deletion reduces bone resorption and abolishes miR-34a regulation.
