Phosphopantothenoylcysteine decarboxylase

phosphopantothenoylcysteine decarboxylase, CoaC, PPCDC
Biosynthesis of coenzyme A (CoA) from pantothenic acid (vitamin B5) is an essential universal pathway in prokaryotes and eukaryotes. PPCDC (EC, one of the last enzymes in this pathway, converts phosphopantothenoylcysteine to 4-prime-phosphopantetheine (Daugherty et al., 2002 [PubMed 11923312]).[supplied by OMIM, Mar 2008] (from NCBI)

Papers on phosphopantothenoylcysteine decarboxylase
Palladium-catalyzed intramolecular aminoacetoxylation of unactivated alkenes with hydrogen peroxide as oxidant.
Liu et al., Shanghai, China. In Org Lett, Apr 2015
Mechanistic study revealed that the addition of di(2-pyridyl) ketone (dpk) ligand was crucial to promote the oxidative cleavage of the C-Pd(II) bond by H2O2 to give the C-OAc bond.
Practical synthesis of anti-β-hydroxy-α-amino acids by Pd(II) -catalyzed sequential C(sp(3) )-H functionalization.
Shi et al., Hangzhou, China. In Chemistry, Mar 2015
The observed diastereoselectivities are strongly influenced by electronic effects of aromatic AAs and steric effects of the linear side-chain AAs, which could be explained by the competition of intramolecular C-OAc bond reductive elimination from Pd(IV) intermediates vs. intermolecular attack by an external nucleophile (AcO(-) ) in an SN 2-type process.
Complex stability and dynamic subunit interchange modulates the disparate activities of the yeast moonlighting proteins Hal3 and Vhs3.
Strauss et al., Stellenbosch, South Africa. In Sci Rep, 2014
Saccharomyces cerevisiae Hal3 and Vhs3 are moonlighting proteins, acting both as inhibitors of the serine/threonine protein phosphatase Ppz1 and as subunits (together with Cab3) of the unique heterotrimeric phosphopantothenoylcysteine decarboxylase (PPCDC) enzyme of Hemiascomycetous yeast.
Novel evidence that crosstalk between the complement, coagulation and fibrinolysis proteolytic cascades is involved in mobilization of hematopoietic stem/progenitor cells (HSPCs).
Ratajczak et al., Louisville, United States. In Leukemia, 2014
Here we provide evidence that during granulocyte colony-stimulating factor- and AMD3100-induced mobilization, not only the ComC but also two other evolutionarily ancient proteolytic enzyme cascades, the coagulation cascade (CoaC) and the fibrynolytic cascade (FibC), become activated.
The Schizosaccharomyces pombe fusion gene hal3 encodes three distinct activities.
Ariño et al., Barcelona, Spain. In Mol Microbiol, 2013
Saccharomyces cerevisiae Hal3 and Vhs3 are moonlighting proteins, forming an atypical heterotrimeric decarboxylase (PPCDC) required for CoA biosynthesis, and regulating cation homeostasis by inhibition of the Ppz1 phosphatase.
Molecular characterization of the heteromeric coenzyme A-synthesizing protein complex (CoA-SPC) in the yeast Saccharomyces cerevisiae.
Schüller et al., Greifswald, Germany. In Fems Yeast Res, 2013
Cab3 also binds to Sis2 and Vhs3 that were previously characterized as subunits of phosphopantothenoylcysteine decarboxylase.
A recessive X-linked mutation causes a threefold reduction of total body zinc accumulation in Drosophila melanogaster laboratory strains.
Missirlis et al., London, United Kingdom. In Febs Open Bio, 2012
Based on a prior report of a threefold difference in zinc accumulation between fumble(1) heterozygous mutants and control fly strains, it was suggested that phosphopantothenoylcysteine decarboxylase might affect zinc status through its effects on vitamin B5 (pantothenate) metabolism.
Functional mapping of the disparate activities of the yeast moonlighting protein Hal3.
Ariño et al., Stellenbosch, South Africa. In Biochem J, 2012
The Saccharomyces cerevisiae Hal3 protein is a moonlighting protein, able to function both as an inhibitory subunit of the Ppz1 protein phosphatase and as a constituent protomer of an unprecedented heterotrimeric PPCDC (phosphopantothenoylcysteine decarboxylase), the third enzyme of the CoA biosynthetic pathway.
Pantethine rescues phosphopantothenoylcysteine synthetase and phosphopantothenoylcysteine decarboxylase deficiency in Escherichia coli but not in Pseudomonas aeruginosa.

Identification of the gene cluster for the dithiolopyrrolone antibiotic holomycin in Streptomyces clavuligerus.
Walsh et al., Boston, United States. In Proc Natl Acad Sci U S A, 2010
Deletions of ORF3488, the NRPS module, and ORF3489, a phosphopantothenoylcysteine decarboxylase homolog, abolished holomycin production both in wild type and in a holomycin-overproducing mutant.
SCAMP5, NBEA and AMISYN: three candidate genes for autism involved in secretion of large dense-core vesicles.
Creemers et al., Leuven, Belgium. In Hum Mol Genet, 2010
Here, we describe the positional cloning of SCAMP5, CLIC4 and PPCDC as candidate genes for autism, starting from a person with idiopathic, sporadic autism carrying a de novo chromosomal translocation.
