Pellino homolog 3

Pellino3, Pellino3a
The protein encoded by this gene is a scaffold protein and an intermediate signaling protein in the innate immune response pathway. The encoded protein helps transmit the immune response signal from Toll-like receptors to IRAK1/TRAF6 complexes. Several transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Jul 2011] (from NCBI)

Papers on Pellino3
The E3 ubiquitin ligase Pellino3 protects against obesity-induced inflammation and insulin resistance.
Moynagh et al., Ireland. In Immunity, 2015
Here we found reduced expression of the E3 ubiquitin ligase Pellino3 in human abdominal adipose tissue from obese subjects and in adipose tissue of mice fed a high-fat diet and showing signs of insulin resistance.
Autophagy-dependent PELI3 degradation inhibits proinflammatory IL1B expression.
von Knethen et al., Frankfurt am Main, Germany. In Autophagy, 2013
Here, we show that PELI3/Pellino3 (pellino E3 ubiquitin protein ligase family member 3), which is an E3 ubiquitin ligase and scaffold protein in TLR4-signaling, is impacted by autophagy in macrophages (MΦ) after LPS stimulation.
Pellino3 ubiquitinates RIP2 and mediates Nod2-induced signaling and protective effects in colitis.
Moynagh et al., Ireland. In Nat Immunol, 2013
Here we found that the E3 ubiquitin ligase Pellino3 was an important mediator in the Nod2 signaling pathway.
Pellino3 targets RIP1 and regulates the pro-apoptotic effects of TNF-α.
Moynagh et al., Ireland. In Nat Commun, 2012
Here we show that deficiency of the ubiquitin E3 ligase, Pellino3, sensitizes cells to TNF-induced apoptosis without inhibiting the NF-κB pathway.
Pellino3 targets the IRF7 pathway and facilitates autoregulation of TLR3- and viral-induced expression of type I interferons.
Moynagh et al., Ireland. In Nat Immunol, 2012
Here we show that genetic ablation of the E3 ubiquitin ligase Pellino3 augmented the expression of type I interferon but not of proinflammatory cytokines in response to TLR3 activation.
OxLDL inhibits LPS-induced IFNβ expression by Pellino3- and IRAK1/4-dependent modification of TANK.
Brüne et al., Frankfurt am Main, Germany. In Cell Signal, 2012
Modified TANK inhibits recruitment of TBK1 (TANK-binding kinase 1) to TRAF3 (TNF receptor associated factor 3) and the subsequent activation of the transcription factor IRF3 (interferon regulatory factor 3). OxLDL stimulates TANK mono-ubiquitination by subsequent activation of IRAK1/4 (interleukin-1 receptor-associated kinases 1 and 4) and Pellino3 downstream of SR-A1 (scavenger receptor-A1).
Mal mediates TLR-induced activation of CREB and expression of IL-10.
Moynagh et al., Ireland. In J Immunol, 2011
We reveal that Mal-mediated activation of CREB is dependent on Pellino3 and TNFR-associated factor 6, because CREB activation is greatly diminished in Pellino3 knockdown cells and TNFR-associated factor 6-deficient cells.
Pellino 3b negatively regulates interleukin-1-induced TAK1-dependent NF kappaB activation.
Li et al., Cleveland, United States. In J Biol Chem, 2008
suggest that Pellino 3b acts as a negative regulator for IL-1 signaling by regulating IRAK degradation through its ubiquitin protein ligase activity [Pellino 3b]
Kinase-active interleukin-1 receptor-associated kinases promote polyubiquitination and degradation of the Pellino family: direct evidence for PELLINO proteins being ubiquitin-protein isopeptide ligases.
Moynagh et al., Ireland. In J Biol Chem, 2007
Recombinant forms of Pellino1 and Pellino2 and both spliced variants of Pellino3 are shown in an in vitro ubiquitination assay to be E3 ligases that catalyze Lys(63)-linked polyubiquitination, with Pellino3 exhibiting the greatest ligase activity.
Pellino3 is a novel upstream regulator of p38 MAPK and activates CREB in a p38-dependent manner.
Moynagh et al., Dublin, Ireland. In J Biol Chem, 2005
Pellino3 is a novel upstream regulator of p38 MAPK that activates CREB in a p38-dependent manner
Pellino3, a novel member of the Pellino protein family, promotes activation of c-Jun and Elk-1 and may act as a scaffolding protein.
Whitehead et al., Philadelphia, United States. In J Immunol, 2003
Two alternatively spliced Pellino3 mRNAs, Pellino3a and Pellino3b, are widely expressed.
