[KRAS and bronchial adenocarcinoma. Between disappointments and hopes].
Toulouse, France. In Rev Mal Respir, 28 Nov 2015
KRAS mutation has the potential to drive the oncogenesis of almost one third of lung adenocarcinomas but it leads to a highly complex proliferation signal involving multiple signaling pathways, explaining the disappointing results of various inhibition strategies of K-ras or its effectors.
Role of retinoids in the prevention and treatment of colorectal cancer.
San Marcos, United States. In World J Gastrointest Oncol, 15 Nov 2015
With respect to colorectal cancer (CRC), several critical mutations dysregulate pathways implicated in progression and metastasis, resulting in aberrant Wnt/β-catenin signaling, gain-of-function mutations in K-ras and phosphatidylinositol-3-kinase/Akt, cyclooxygenase-2 over-expression, reduction of peroxisome proliferator-activated receptor γ activation, and loss of p53 function.
RAS-MAPK dependence underlies a rational polytherapy strategy in EML4-ALK-positive lung cancer.
San Francisco, United States. In Nat Med, Sep 2015
Reactivation of the MAPK pathway via either a gain in the number of copies of the gene encoding wild-type K-RAS (KRAS(WT)) or decreased expression of the MAPK phosphatase DUSP6 promoted resistance to ALK inhibitors in vitro, and each was associated with resistance to ALK inhibitors in individuals with EML4-ALK-positive lung adenocarcinoma.

The stability and cellular distribution of Nrf2 is tightly controlled by its inhibitory binding protein Kelch-like ECH-associated protein 1. Nrf2 signalling is also regulated by posttranslational, transcriptional, translational and epigenetic mechanisms, as well as by other protein partners, including p62, p21 and IQ motif-containing GTPase activating protein 1.
