Insulin-like growth factor 2 receptor

Igf2r, insulin-like growth factor II receptor, IGF-II receptor, CI-MPR
The protein encoded by this gene is a receptor for both, insulin-like growth factor 2 (IGF2) and mannose 6-phosphate (M6P). The IGF2 and M6P binding sites are located on different segments of the receptor. This receptor functions in the intracellular trafficking of lysosomal enzymes, the activation of transforming growth factor beta, and the degradation of IGF2. While the mouse Igf2r gene shows exclusive expression from the maternal allele, imprinting of the human IGF2R gene appears to be polymorphic, with only a minority of individuals showing expression from the maternal allele. GCs are required for the differentiation of mesenchymal stem cells to bone cells but in pharmacologic doses they reduce osteoblast proliferation and differentiation, they partially inhibit the production of IL-1 and –6 and inhibit the bone resorbing activity of these cytokines, they interfere with the receptor activator of the nuclear factor-κB ligand (RANKL or TRANCE)-osteoprotegerin (OPG) axis and GCs inhibit IGF-II receptor expression in osteoblasts and decrease the expression of IGFBP-3, -4, and -5 in osteoblasts.
