Hect domain and RLD 3

HERC3
HERC3 belongs to the HERC family of ubiquitin ligases, all of which contain a HECT domain and at least 1 RCC1 (MIM 179710)-like domain (RLD). The 350-amino acid HECT domain is predicted to catalyze the formation of a thioester with ubiquitin before transferring it to a substrate, and the RLD is predicted to act as a guanine nucleotide exchange factor for small G proteins (Hochrainer et al., 2005 [PubMed 15676274]).[supplied by OMIM, Mar 2008] (from NCBI)

Papers on HERC3
The ubiquitin ligase HERC3 attenuates NF-κB-dependent transcription independently of its enzymatic activity by delivering the RelA subunit for degradation.
New
Anrather et al., New York City, United States. In Nucleic Acids Res, Dec 2015
Here, we identify the HECT-domain ubiquitin ligase HERC3 as novel negative regulator of NF-κB activity.
Epigenetic control of alternative mRNA processing at the imprinted Herc3/Nap1l5 locus.
Oakey et al., London, United Kingdom. In Nucleic Acids Res, 2012
Nap1l5 is a retrogene expressed from the paternally inherited allele, is situated within an intron of a 'host' gene Herc3, and overlaps a CpG island that is differentially methylated between the parental alleles.
Frameshift mutations of ubiquitination-related genes HERC2, HERC3, TRIP12, UBE2Q1 and UBE4B in gastric and colorectal carcinomas with microsatellite instability.
GeneRIF
Lee et al., In Pathology, 2011
we found somatic mutations of HERC2, HERC3, TRIP12, UBE2Q1 and UBE4B genes in gastric carcinoma and colorectal carcinomas with microsatellite instability
Progressive Purkinje cell degeneration in tambaleante mutant mice is a consequence of a missense mutation in HERC1 E3 ubiquitin ligase.
Rosa et al., Paris, France. In Plos Genet, 2009
In humans, six HERC genes have been reported which encode two subgroups of HERC proteins: large (HERC1-2) and small (HERC3-6).
Highly homologous HERC proteins localize to endosomes and exhibit specific interactions with hPLIC and Nm23B.
Lipp et al., Vienna, Austria. In Cell Mol Life Sci, 2008
We demonstrate interaction of HERC3 with the ubiquitin-like proteins hPLIC-1 and hPLIC-2 and we establish interaction of HERC5 with the metastasis suppressor Nm23B.
Expression patterns of the novel imprinted genes Nap1l5 and Peg13 and their non-imprinted host genes in the adult mouse brain.
Wilkinson et al., Cambridge, United Kingdom. In Gene Expr Patterns, 2004
Nap1l5 and Peg13 are located, respectively, within the introns of the non-imprinted genes Herc3 and the Tularik1 (T1)/KIAA1882 homologue in two separate microimprinted domains on mouse chromosomes 6 and 15.
Specific association between alcohol intake, high grade of differentiation and 4q34-q35 deletions in hepatocellular carcinomas identified by high resolution allelotyping.
Zucman-Rossi et al., Paris, France. In Oncogene, 2002
Seven putative candidate genes located in 4q22, DAPP1, BMPR1B, PKD2, HERC3, SMARCAD1, CEB1 and ENH were screened for mutations but no somatic alterations were identified.
