F-box and leucine-rich repeat protein 21

This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class and, in addition to an F-box, contains 6 tandem leucine-rich repeats. The amino acid sequence of this protein is highly similar to that of f-box and leucine-rich repeat protein 3A. Comparisons of this gene to orthologous sequences suggest that it may be a pseudogene, and may no longer express a functional protein.[provided by RefSeq, Aug 2009] (from NCBI)

Investigation of genetic variants in ubiquitin enzyme genes involved in the modulation of neurodevelopmental processes: a role in schizophrenia susceptibility?
Fernandez-Enright et al., Wollongong, Australia. In Genet Res (camb), 2013
We have examined single nucleotide polymorphisms (or SNPs) within three genes from the ubiquitin protein system: the ubiquitin conjugating enzyme E2D1 (UBE2D1) gene, the E3 SUMO-protein ligase protein inhibitor of activated STAT 2 (PIAS2) gene, and the E3 ubiquitin ligase F-box and leucine-rich repeat protein 21 (FBXL21) gene, in a Caucasian case-control population for schizophrenia.
FBXL21 regulates oscillation of the circadian clock through ubiquitination and stabilization of cryptochromes.
Fukada et al., Tokyo, Japan. In Cell, 2013
Here, we found that FBXL21 also ubiquitinates CRYs but counteracts FBXL3.
Competing E3 ubiquitin ligases govern circadian periodicity by degradation of CRY in nucleus and cytoplasm.
Takahashi et al., Dallas, United States. In Cell, 2013
Cloning of a short-period circadian mutant, Past-time, revealed a glycine to glutamate missense mutation in Fbxl21, an F-box protein gene that is a paralog of Fbxl3 that targets the CRY proteins for degradation.
FBXL21 association with schizophrenia in Irish family and case-control samples.
Kendler et al., Richmond, United States. In Am J Med Genet B Neuropsychiatr Genet, 2008
Using both family and case-control samples, consistent associations in and around FBXL21 gene suggested that FBXL21 may be associated with schizophrenia in the Irish samples.
Implication of the F-Box Protein FBXL21 in circadian pacemaker function in mammals.
Hazlerigg et al., Aberdeen, United Kingdom. In Plos One, 2007
Our data show that Fbxl3 function is conserved and further reveal that its closest homologue, the F-Box protein Fbxl21, also binds to CRY1 which impairs its repressive action towards the transcriptional activators CLOCK/BMAL1.
